The intracellular fatty acid-binding proteins (FABPs) belong to a multigene family with nearly twenty identified members. FABPs are divided into at least three distinct types, namely the hepatic-, intestinal- and cardiac-type. They form 14-15 kDa proteins and are thought to participate in the uptake, intracellular metabolism and/or transport of long-chain fatty acids. They may also be responsible in the modulation of cell growth and proliferation. Intestinal fatty acid-binding protein 2 gene contains four exons and is an abundant cytosolic protein in small intestine epithelial cells. This gene has a polymorphism at codon 54 that identified an alanine-encoding allele and a threonine-encoding allele. Thr-54 protein is associated with increased fat oxidation and insulin resistance. CLP36, also known as PDLIM1, CLIM1 or Elfin, is a 329 amino acid cytoplasmic protein that associates with actin stress fibers at the cytoskeleton. Expressed at high levels in skeletal muscle and heart and at lower levels in colon, small intestine, spleen, lung, placenta, kidney, liver, thymus and pancreas, CLP36 functions as a cytoskeletal protein that is thought to act as an adaptor, bringing target proteins to the cytoskeleton. Specifically, CLP36 interacts with Clik1 (-actinin-1, thereby facilitating the association of Clik1 with actin stress fibers. CLP36 contains one PDZ domain and one LIM zinc-binding domain through which it conveys its protein-protein binding capabilities. Human CLP36 shares 88% sequence similarity with its rat counterpart, suggesting a conserved function between species. The progression of cells through the cell cycle is regulated by a family of proteins designated cyclin-dependent kinases (Cdks). Sequential activation of individual members of this family and their consequent phosphorylation of critical substrates promotes orderly progression through the cell cycle. Multiple proteins are encoded by the tumor suppressor gene CDKN2A (MTS1/ p16INK4a) via translation of alternate reading frames, resulting in the pro- duction of the p19 ARF protein in mice and the p14 ARF protein in humans. p14 ARF induces an increase in MDM2 and p21 levels and leads to cell cycle arrest in both G1 and G2/M. p14 ARF is negatively regulated by p53 and is known to bind directly to MDM2. CDKN2A also encodes the mitotic protein p16, which binds to and inhibits the Cdk4/cyclin D complex. DNA methylation is the major modification of eukaryotic genomes and plays an essential role in mammalian development. Human proteins MECP2, MBD1, MBD2, MBD3, and MBD4 comprise a family of nuclear proteins related by the presence in each of a methyl-CpG binding domain (MBD). Each of these proteins, with the exception of MBD3, is capable of binding specifically to methylated DNA. MECP2, MBD1 and MBD2 can also repress transcription from methylated gene promoters. MBD1 has also been predicted to be involved in the development and progression of tumors, and it has been shown that MBD1 is significantly upregulated in pancreatic cancer tissues when compared with surrounding normal tissues. Geminin is a nuclear protein that regulates the initiation of DNA replication during the cell cycle. DNA replication requires the coordinated association of Cdc6 and minichromosome maintenance (MCM) proteins with chromatin. Geminin blocks this assembly of the MCM into the prereplication complex and, in turn, prevents replication from occurring. Expression of Geminin fluctuates throughout the cell cycle with Geminin levels lowest at G1. Throughout S, G2 and M phases, Geminin levels are consistently elevated followed by a decrease during mitosis. The initiation of DNA replication is dependent on the degradation of Geminin during mitosis and the absence of Geminin throughout G1 phase.
